A. ferrooxidans have a great scientific interest being involved in iron oxidation; therefore, an accurate growth medium for viable cell detection is very much needed. The aim of the present study is to compare different techniques and media such as: membrane filtration method, spread plate enumeration used for growth and recovery of these microorganisms and possibly improve it. In the present study the traditional solidifying agent (agar or agarose) will be replaced with Gelrite. The effect of parameters such as: Gelrite concentration and other nutrient medium components, age of solid media, initial concentration of bacterial cells, incubation period, etc., will be tested for the recovery ability. The expected result is to find the most suitable solid nutrient medium for isolation and maximal growth of Thiobacillus ferrooxidans to be enumerated from environmental samples.
